Ensign, J. C. (University of Wisconsin, Madison), and R. S. Wolfe. Characterization of a small proteolytic enzyme which lyses bacterial cell walls. J. Bacteriol. 91:524-534. 1966.-An enzyme isolated from a myxobacter possesses both cell-wall lytic and proteolytic activity. The enzyme has been purified over 600-fold and is electrophoretically homogeneous upon cellulose acetate at several pH values and upon polyacrylamide gel columns. A single peak was obtained upon ultracentrifugation and density gradient centrifugation. Based upon Sephadex gel filtration, a molecular weight of 8,700 was determined for the enzyme. Albumin and casein were extensively degraded by the enzyme, with approximately one-third of the peptide bonds present in these proteins being hydrolyzed. The enzyme lyses cell walls by hydrolyzing peptide bonds in the glycosaminopeptide.

Validation of a Simple and Rapid Method for Assessment of Intracellular Bacterial Asparaginase

L-Asparaginase has remarkable properties which make it useful in dual pharmaceutical and food industries. In this study, simple and advantageous method has been validated for rapid and precise determination of intracellular L-Asparaginasein bacterial species. A suspension of bacterial cells was used instead ofcell extract and incubated by substrate (asparagine) after simple wash and centrifugati...

Bacterial Expression and Functional Characterization of A Naturally Occurring Exon6-less Preprochymosin cDNA

Chymosin (Rennin EC 3.4.23.4), an aspartyl proteinase, is the major proteolytic enzyme in the fourthstomach of the unweaned calf, and it is formed by proteolytic activation of its zymogene, prochymosin.Following the cloning of synthesized cDNAs on mRNA pools extracted from the mucosa of the calf fourthstomach, we have identified an alternatively spliced form of preprochymosin ...

Lytic enzyme produced by Myxococcus xanthus.

Hart, Beth A. (Cornell University, Ithaca, N.Y.), and Stanley A. Zahler. Lytic enzyme produced by Myxococcus xanthus. J. Bacteriol. 92:1632-1637. 1966.-Strain FBa of Myxococcus xanthus releases into its culture medium an enzyme capable of lysing Micrococcus lysodeikticus cells and of releasing N-acetyl amino sugars from their cell walls. The lysin is stable at pH values near neutrality and at t...

Characterization of a proteolytic enzyme from Lachesis muta venom (Serpentes: Viperidae).

A proteolytic enzyme from L. muta stenophrys was isolated by gel filtration on Bio Gel P-100 followed by FPLC on MONO S column. The enzyme exhibited proteolytic activity toward casein, hemoglobin and fibrinogen with a pH optimum around 10. The activity was inhibited by EDTA while trypsin inhibitors were not inhibitory. It is a glycoprotein, Mr 14 kDa with a high content of Asp, Glu, and Leu res...

Purification and Partial Characterization of a Thrombin-Like Enzyme (AH144) from Venom of Iranian Snake Agkistrodon Halys

The snake venom´s thrombin-like enzymes comprise a number of serine proteases, which are functionally and structurally related to thrombin. Purification and partial characterization of a thrombin-like enzyme from the venom of the Iranian snake, Agkistrodon halys, was the aim of this study. Purification was carried out by a combination of variety of chromatographic methods that included: gel...
